1. 서론

1960년대부터 국제원자력기구(International Atomic Energy Agency, IAEA)는 강수 동위원소 글로벌 네트워크(Global Network for Isotopes in Precipitation)를 운영하여 전세계 강수의 수소(H) 및 산소(O) 안정동위원소비를 측정하였고 강수의 위도, 고도 및 계절 등 다양한 인자를 이용해 강수 및 수증기 기원과 대기 순환 과정을 해석하였다(Craig, 1961; Rozanski et al., 1993). 최근에는 원산지 및 진위 판별을 위해 병입수(bottled water), 와인 및 음료수 등을 대상으로 수소 및 산소동위원소를 분석함으로써 안정동위원소의 연구 범위가 급격히 확장되었다(Roßmann et al., 1999; Bowen et al., 2005). 병입수의 화학, 수소 및 산소동위원소 조성은 지하수 특성, 기원 추적, 가뭄 및 지질학적 활동(지진, 화산)의 모니터링 등 수리지질학적 연구에서도 매우 중요하게 활용되고 있다(Tsunogai and Wakita, 1995; Chiogna et al., 2018; Lee et al., 2021).

물의 안정동위원소비는 전통적으로 안정동위원소 질량분석기(isotope ratio mass spectrometry, IRMS)를 이용해 측정되었으나, 최근에는 레이저(laser)가 결합된 동위원소비 적외선 분광분석기(isotope ratio infrared spectroscopy, IRIS)도 많이 사용한다. 후자는 비교적 간단한 시료 전처리, 장비 유지 보수 및 현장 활용이 가능한 장점이 있지만, 유기물 함량이 높거나 분광 간섭의 영향을 줄 수 있는 용매가 포함된 시료 등의 IRIS 분석 결과는 IRMS의 결과와 상당한 차이를 보이기도 한다(Brand et al., 2009; West et al., 2010; Zhao et al., 2011). 반면, 전처리 라인을 통해 순수하게 분리된 시료가스와 표준가스를 번갈아 분석할 수 있는 이중 시료 도입부(dual-inlet mode) 또는 혼합가스와의 물리적 평형을 이용하는 가스벤치(gasbench)가 결합된 IRMS 분석법은 극미량 시료나 용존물(유기물, 고염도)이 포함된 시료도 분석이 가능하며 상대적으로 높은 정밀도를 가진 분석 결과를 얻을 수 있다(Wassenaar et al., 2018; Terzer-Wassmuth et al., 2020).

물의 산소동위원소비는 일반적으로 가스벤치를 이용한 이산화탄소 평형법(CO₂ equilibrium method)을 적용하여 측정하며, 니켈(Ni)/탄소(C) 가열로와 오산화아이오딘(I₂O₅)을 통해 CO₂를 생성시킨 후 측정하기도 한다(Santrock and Hayes, 1987; Costinel et al., 2009). 물의 수소동위원소비는 아연(Zn), 크롬(Cr) 및 망간(Mn) 등 다양한 화합물의 조합으로 환원시키거나 Ni/C 혼합물이 포함된 가열로를 이용하여 H₂ 가스를 생성시켜 측정한다(Coleman et al., 1982; Tanweer and Han, 1996; Morrison et al., 2001; Gehre and Strauch, 2003). 또한, 백금 촉매(platinum catalyst)와 수소 혼합 가스를 이용한 수소 평형법(H₂ equilibrium method)도 활용되고 있다. 앞서 언급한 사례 가운데 평형법을 제외한 측정 방법은 시료에 용해된 염에 민감한 경향이 있으며 염은 금속 가열로와 시료 간의 반응을 완전히 또는 부분적으로 방해한다(Midwood and McGaw, 1999). 최근에는 원소분석기(elemental analyzer, EA) 내 유리 탄소 튜브(glassy carbon tube)와 고온(≥1,400℃)을 이용해 간편하게 물을 분해하는 기술이 많이 이용되고 있다(Sharp et al., 2001; Gehre et al., 2004; Magozzi et al., 2020). 이 방법은 평형법을 이용한 분석 결과보다 유사하거나 약간 낮은 정밀도를 가지나 앞서 언급한 것보다 분석 전처리가 단순하고 산소와 수소동위원소비를 동시에 측정할 수 있어 분석 시간이 짧다. 이번 연구에서는 최근 한국지질자원연구원에 설치된 고감도(high sensitivity, HS) IRMS 시스템을 소개하고, IRMS와 고온 연소 EA (high-temperature conversion EA, HTC-EA)를 이용하여 천연수(natural water)와 병입수의 산소 및 수소동위원소 분석을 수행하였다.

2. 고감도 안정동위원소 질량분석 시스템

한국지질자원연구원 지질자원분석센터에 설치된 HS-IRMS 시스템은 HS-IRMS (253 Plus, Thermo Fisher Scientific, Bremen, Germany), EA (EA IsoLink CNS and EA IsoLink OH, Thermo Fisher Scientific, Bremen, Germany), 가스벤치(GasBench II, Thermo Fisher Scientific, Bremen, Germany) 및 가스 선 농축장치(PreCon, Thermo Fisher Scientific, Bremen, Germany) 등으로 구성되어 있다(Fig. 1). 연속 흐름(continuous flow, CF) 가스 제어장치(ConFlo IV, Thermo Fisher Scientific, Bremen, Germany)를 이용해 IRMS와 전처리 장치를 연결하고 전용 프로그램을 통해 자동제어 및 연속 흐름 방식의 분석이 가능하다. 이뿐만 아니라 이중 시료 도입부가 질량분석기 내에 탑재되어 있어 극미량 시료의 동위원소비를 정밀하게 측정할 수 있다. HS-IRMS의 가속전압은 약 10 kV이며, 보편적으로 사용하는 ~5 kV IRMS보다 분해능이 높고 감도가 좋다. 7개의 패러데이 컵(Faraday cup) 중 2개는 HD 전용 검출기로 전자석(magnet) 앞쪽에 위치하여 수소동위원소 분석에 최적화하였다(Fig. 1 and Supplemental table 1). 2대의 EA를 이용해 고체와 액체 시료에서 탄소, 질소(N), 황(S), 산소 및 수소를 기화하여 분리하거나 추출할 수 있다. 가스벤치와 가스 선 농축장치를 이용해 탄산염암의 탄소 및 산소동위원소 동시 분석과 탈질 박테리아법을 이용한 질산염의 질소 및 산소동위원소의 동시 분석도 가능하다. 총 9종의 참고, 운반, 산화 및 평형 가스를 설치하였고, 참고 가스는 대부분 고순도(≥99.999%)를 사용하였다(Supplemental table 2). 모든 참고 가스는 연속 흐름 가스 제어장치를 통해 자동 도입된다. 구축된 시스템으로 미량의 유기물, 무기물 및 물 시료 등의 산소, 수소, 탄소, 질소 및 황동위원소 분석이 가능하다.

3. 시료 및 분석 방법

3.1. 분석 물질

이번 연구에서 사용한 분석법의 신뢰성 검증을 위해 국제 숙련도 시험인 2020 WICO (Water Isotope InterComparison)에 참가하여 IAEA로부터 6종의 천연수 시료를 제공받았다. 시험 물질 중 하나인 OH-30은 OH-26과 동일한 수돗물(Vienna tap water)로 분석 재현성을 검증하기 위한 블라인드(blind) 시료이다(Wassenaar et al., 2021). 신뢰성 검증 후, 물의 수소 및 산소동위원소 분석을 위해 병입수 14종(국내 9종, 해외 5종)을 온라인 및 오프라인 마트에서 2021년 상반기에 구매하였다. 수원지가 백두산(중국)인 2종은 해외 병입수로 분류하였다. 국내 병입수 중 7종은 암반 대수층(선캠브리아기 변성퇴적암, 선캠브리아기 편마암, 백악기 화강암 및 신생대 화산암) 지하수이며, 나머지 2종은 해양 심층수이다. 해외 병입수의 원수원은 (화산) 암반 대수층 및 빙하수이다. 14종에 대한 자세한 정보는 Supplemental table 3에 나타내었다. 국제 물 표준물질인 VSMOW2 (Vienna Standard Mean Ocean Water 2), SLAP2 (Standard Light Antarctic Precipitation 2) 및 USGS46 (Ice Core Water)도 함께 분석하였다.

3.2. 물의 수소 및 산소동위원소 분석

HTC-EA를 이용한 물의 안정동위원소 분석은 혼합 가스 평형법을 이용한 분석에 비하여 시료 전처리가 간단하다. 물 시료 채취 시 0.45 μm 크기의 필터를 이용하여 불순물 및 부유물 등을 걸러 대기와 접촉되지 않게 용기에 가득 채워 냉장 보관하였다. 분석 전, 시료와 표준물질은 실험실의 대기와 온도 평형을 이루도록 3시간 이상 방치하였다. 시료 분체를 위해 2.5 mL 피펫(pipette, Eppendorf, Germany)과 별도의 세척이 필요 없는 피펫팁(“PCRclean” tip, Eppendorf, Germany)을 사용하였으며, 2 mL의 유리 용기(clear Target DP ID^TM glass vial, Thermo Fisher Scientific, USA)에 가득 채웠다. 마이크로 시린지(syringe) 세척을 위해 에탄올(ethyl alcohol, GR급), 아세톤(acetone, GR급) 및 초순수(Millipore Milli-Q, 18.2 MΩ·cm^-1)를 사용하였다.

천연수와 병입수의 수소 및 산소동위원소 분석을 위하여 액체 오토 샘플러의 시린지를 통하여 1uL의 시료를 HTC-EA에 주입하였다. 시료를 주입하기 전과 후에 초순수로 충분히 세척하여 연소로 인한 미세 그을음 및 메모리 효과(memory effect)를 제거하였다. 연소 온도는 1400℃이며 bottom-feed adaptor 방식을 이용하여 운반 가스(He)를 공급하였다. 수소와 산소 가스를 추출하기 위해 글래시 카본칩이 충진된 글래시 탄소 튜브와 세라믹 튜브를 반응로(reactor)로 사용하였다. EA로 분리된 시료의 수소 및 산소 가스는 모세관(capillary tube)을 통해 연속 흐름 가스 제어장치를 거쳐 참고 가스(H₂와 CO)와 교차로 IRMS에 도입된다. EA를 통해 도입되는 H₂와 CO 가스는 EA 내부의 GC 오븐(gas chromatography oven)을 거쳐 도입되므로, 각 가스의 도입 시기에 맞춰 자기장의 세기를 변화하는 피크 점프(peak jump)를 수행하여 두 종류의 가스를 연속 분석하였다. 자세한 HTC-EA와 IRMS의 분석 조건은 Supplemental table 4에 나타내었다.

분석시료는 5회 이상 측정하였고, 불완전 연소, 메모리 효과 및 시린지 오작동 등으로 발생한 결과는 계산과정에서 제외하였다. 오차는 표준편차(1 standard deviation, 1 SD)로 나타내었다. 측정값의 보정을 위해 두 종의 국제 표준물질인 VSMOW2와 SLAP2을 이용하였으며 이점 선형 보정법(two-point linear normalization)으로 질량 분별(instrumental mass fractionation)을 보정하였다(Nelson, 2000; IAEA 2009). ²H/¹H 및 ¹⁸O/¹⁶O 비는 VSMOW로 규격화하여 델타 표시법(δ, delta notation)으로 나타내었다(Coplen, 1988). 신뢰성 확보를 위해 작업 표준물질인 USGS46를 반복 분석하였다. 측정된 수소와 산소동위원소비는 각각 –235.5 ± 1.1 ‰(1 SD, n= 7) 및 –29.81 ± 0.07 ‰(1 SD, n= 7)이며, 참고 값과 오차범위 내에서 일치한다(Coplen et al., 2014). 추가적인 신뢰성 검증을 위해 선별된 병입수 시료(10종)의 산소동위원소비는 국립해양생물자원관에 설치된 3kV의 IRMS (Delta V Plus, Thermo Fisher Scientific, Bremen, Germany)와 가스벤치를 이용하여 측정하였다.

4. 결과 및 토의

4.1. 2020 WICO

WICO는 IAEA에서 주관하는 물 동위원소 분석 능력을 검증하는 국제 숙련도 시험으로 4년마다 개최된다. 2020 WICO는 2016 WICO 시험 물질보다 염도가 낮고 비교적 순수한 천연수의 δ¹⁸O 및 δD 값을 비교하여 실험실의 분석 능력을 검증하였다(Wassenaar et al., 2021). 총 6종의 천연수에 대하여 δ¹⁸O와 δD 값을 분석하여 결과를 보고하였다. 측정한 천연수 시료는 IAEA에서 제시한 참고 값과 오차범위 내에서 일치하며, z-검정(z-test) 및 ζ-검정 값이 각각 –0.67~+1.10 및 –0.99~+1.00의 범위로 ‘만족(satisfactory)’ 판정을 받았다(Fig. 2 and Supplemental material 1). 2020 WICO 테스트의 자세한 내용은 Wassenaar et al. (2021)에서 찾아볼 수 있다.

4.2. 병입수의 안정동위원소 조성

14종의 병입수의 안정동위원소 분석 결과는 Table 1에 나타내었다. HTC-EA를 이용한 산소동위원소 분석 결과는 CO₂ 평형법을 이용한 분석 결과와 오차범위 내에서 일치한다(Supplemental table 5). 원수원이 암반 대수층인 국내 병입수 7종의 수소와 산소동위원소 조성 범위는 –61~–43 ‰ 및 –8.9~–7.1 ‰이다. 2종의 국내 해양 심층수의 평균 수소와 산소동위원소 조성은 0.2 ± 0.2 ‰(1 SD) 및 –0.12 ± 0.12 ‰(1 SD)이다. 이는 VSMOW2의 안정동위원소 조성과 유사하다. 수원지가 백두산인 시료를 포함한 수입 병입수의 수소와 산소동위원소 조성은 – 100~–41 ‰ 및 –14.1~–6.5 ‰이다. 수원지가 피지섬인 BT-57 시료를 제외한 나머지는 국내 병입수보다 낮은 안정동위원소 조성을 보였다. 비록 BT-54의 수소동위원소비가 약간 낮으나, BT-51, BT-57 및 BT-70 시료의 안정동위원소 조성은 기존에 보고된 수원지가 인접한 지역에서 채취한 시료 값과 오차범위 내에서 일치한다(Bong et al., 2009; Kim et al., 2012; Lee et al., 2021). 해양 심층수를 제외한 국내 병입수와 원수원이 백두산인 시료의 분석 결과에서 경도가 높을수록 낮은 안정동위원소 조성을 보이며, 안정동위원소비가 지구 순환 수선(Global Meteoric Water Line: GMWL, Craig, 1961)과 지역 순환 수선(Local Meteoric Water Line: LMWL, Gautam et al., 2020) 위에 도시된다(Fig. 3). 이를 통해, 국내 병입수의 안정동위원소 조성은 강수의 조성을 반영하며, 해외 병입수와 구별이 가능하다.

5. 결론

국제 숙련도 시험인 2020 WICO를 통해 분석 신뢰성을 검증받은 후, KIGAM에 설치된 HS-IRMS와 HTC-EA를 이용하여 천연수와 병입수의 안정동위원소 분석을 수행하였다. 평형법을 적용한 국내외 병입수의 안정동위원소 분석 결과와 교차 비교함으로써 신뢰성을 재검증하였다. 이번 연구를 통해 HTC-EA를 활용한 분석법은 강수, 지하수 및 병입수 등 유기물이 적고 염도가 낮은 깨끗한 물 시료의 안정동위원소 분석에 유용하게 활용될 것으로 기대된다. 현재, 한국지질자원연구원의 온라인 분석 시스템(https://gac.kigam.re.kr)을 통해 위 분석법을 이용한 물 시료의 안정동위원소 분석 서비스를 제공하고 있다. 향후 다양한 온라인 전처리 장치를 활용한 유기물 및 무기물의 안정동위원소 분석 항목을 확대할 계획이다.
